Thermal treatment of magnesium particles in polylactic acid polymer films elicits the expression of osteogenic differentiation markers and lipidome profile remodeling in human adipose stem cells
The efficacy of polylactic acid (PLA)/Magnesium (Mg)-based materials for driving stem cells toward bone tissue engineering applications requires specific Mg surface properties to modulate the interface of stem cells with the film. Here, we have developed novel PLA/Mg-based composites and explored their osteogenic differentiation potential on human adipose stem cells (hASCs). Mg-particles/polymer interface was improved by two treatments: heating in oxidative atmosphere (TT) and surface modification with a compatibilizer (PEI). Different contents of Mg particles were dispersed in PLA and composite surface and bulk properties, protein adsorption, stem cell-PLA/Mg interactions, osteogenic markers expressions, and lipids composition profile were evaluated. Mg particles were uniformly distributed on the surface and in the bulk PLA polymer. Improved and modulated particle-polymer adhesion was observed in Mg particle-treated composites.
After 21 days in canonical growth culture conditions, hASCs on PLA/MgTT displayed the highest expression of the general osteogenic markers, RUNX2, SSP1, and BGLAP genes, Alkaline Phosphatase, type I Collagen, Osteopontin, and Calcium deposits. Moreover, by LC/MS QTOF mass-spectrophotometry lipidomic analysis, we found in PLA/MgTT-cells, for the first time, a remodeling of the lipid classes composition associated with the osteogenic differentiation. We ascribed these results to MgTT characteristics, which improve Mg availability and composite osteoinductive performance.This work has been carried out within the M-ERANET Programme: Project POLYMAGIC. Four aspects of self-image close to death at home
Living close to death means an inevitable confrontation with one's own existential limitation. In this article, we argue that everyday life close to death embodies an identity work in progress. We used a narrative approach and a holistic-content reading to analyze 12 interviews conducted with three persons close to death. By illuminating the unique stories and identifying patterns among the participants’ narratives, we found four themes exemplifying important aspects of the identity work related to everyday life close to death. Two of the themes, named “Inside and outside of me” and “Searching for togetherness,” represented the core of the self-image and were framed by the other themes, “My place in space” and “My death and my time.” Our findings elucidate the way the individual stories moved between the past, the present, and the future. This study challenges the idea that everyday life close to impending death primarily means limitations. The findings show that the search for meaning, new knowledge, and community can form a part of a conscious and ongoing identity work close to death
The Neurokinin 1 Receptor Antagonist, Ezlopitant, Reduces Appetitive Responding for Sucrose and Ethanol
Abstract Background: The current obesity epidemic is thought to be partly driven by over-consumption of sugar-sweetened diets and soft drinks. Loss-of-control over eating and addiction to drugs of abuse share overlapping brain mechanisms including changes in motivational drive, such that stimuli that are often no longer ‘liked’ are still intensely ‘wanted’ [7,8]. The neurokinin 1 (NK1) receptor system has been implicated in both learned appetitive behaviors and addiction to alcohol and opioids; however, its role in natural reward seeking remains unknown. Methodology/Principal Findings: We sought to determine whether the NK1-receptor system plays a role in the reinforcing properties of sucrose using a novel selective and clinically safe NK1-receptor antagonist, ezlopitant (CJ-11,974), in three animal models of sucrose consumption and seeking. Furthermore, we compared the effect of ezlopitant on ethanol consumption and seeking in rodents. The NK1-receptor antagonist, ezlopitant decreased appetitive responding for sucrose more potently than for ethanol using an operant self-administration protocol without affecting general locomotor activity. To further evaluate the selectivity of the NK1-receptor antagonist in decreasing consumption of sweetened solutions, we compared the effects of ezlopitant on water, saccharin-, and sodium chloride (NaCl) solution consumption. Ezlopitant decreased intake of saccharin but had no effect on water or salty solution consumption. Conclusions/Significance: The present study indicates that the NK1-receptor may be a part of a common pathway regulating the self-administration, motivational and reinforcing aspects of sweetened solutions, regardless of caloric value, and those of substances of abuse. Additionally, these results indicate that the NK1-receptor system may serve as a therapeutic target for obesity induced by over-consumption of natural reinforcers
Spatial Learning and Action Planning in a Prefrontal Cortical Network Model
The interplay between hippocampus and prefrontal cortex (PFC) is fundamental to
spatial cognition. Complementing hippocampal place coding, prefrontal
representations provide more abstract and hierarchically organized memories
suitable for decision making. We model a prefrontal network mediating
distributed information processing for spatial learning and action planning.
Specific connectivity and synaptic adaptation principles shape the recurrent
dynamics of the network arranged in cortical minicolumns. We show how the PFC
columnar organization is suitable for learning sparse topological-metrical
representations from redundant hippocampal inputs. The recurrent nature of the
network supports multilevel spatial processing, allowing structural features of
the environment to be encoded. An activation diffusion mechanism spreads the
neural activity through the column population leading to trajectory planning.
The model provides a functional framework for interpreting the activity of PFC
neurons recorded during navigation tasks. We illustrate the link from single
unit activity to behavioral responses. The results suggest plausible neural
mechanisms subserving the cognitive “insight” capability originally
attributed to rodents by Tolman & Honzik. Our time course analysis of neural
responses shows how the interaction between hippocampus and PFC can yield the
encoding of manifold information pertinent to spatial planning, including
prospective coding and distance-to-goal correlates
Nef divisors for moduli spaces of complexes with compact support
In [BM14b], the first author and Macr\`i constructed a family of nef divisors
on any moduli space of Bridgeland-stable objects on a smooth projective variety
X. In this article, we extend this construction to the setting of any separated
scheme Y of finite type over a field, where we consider moduli spaces of
Bridgeland-stable objects on Y with compact support. We also show that the nef
divisor is compatible with the polarising ample line bundle coming from the GIT
construction of the moduli space in the special case when Y admits a tilting
bundle and the stability condition arises from a \theta-stability condition for
the endomorphism algebra.
Our main tool generalises the work of Abramovich--Polishchuk [AP06] and
Polishchuk [Pol07]: given a t-structure on the derived category D_c(Y) on Y of
objects with compact support and a base scheme S, we construct a constant
family of t-structures on a category of objects on YxS with compact support
